FORMULASI NANOPARTIKEL RIBOSOME-INACTIVATING PROTEIN Mirabilis jalapa L. (RIP MJ) TERTARGET MENGGUNAKAN KITOSAN RANTAI PENDEK - PEKTIN TERKONJUGASI ANTIBODI ANTI-EPCAM DAN UJI SITOTOKSIK PADA SEL KANKER PAYUDARA

Ribosome Inactivating Proteins (RIPs) merupakan protein dari tanaman yang memiliki kemampuan untuk mendepurinasi RNA. RIPs yang diisolasi dari daun Mirabilis jalapa L. memiliki efek sitotoksik yang lebih tinggi pada sel malignan (T47D dan MCF7) daripada pada sel normal. Kitosan nanopartikel sudah banyak digunakan pada aplikasi penghantaran protein terapetik. Namun, studi mengenai penghantaran protein dengan nanopartikel kitosan yang dimodifikasi dengan antibodi target spesifik EpCAM pada jaringan tumor belum dilakukan. Tujuan dari penelitian ini adalah untuk mendapatkan bentuk RIP MJ dalam nanopartikel terkonjugasi antibodi antiEpCAM, mengetahui karakteristik nanopartikel yang dihasilkan dan mengetahui efek sitotoksik nanopartikel pada sel T47D. RIPs dalam nanopartikel kitosan (RIPs nano kitosan-pektin) diformulasikan menggunakan kitosan rantai pendek dan pektin dengan metode polielektrolik kompleks, kemudian dikonjugasikan dengan antibodi antiEpCAM dikonjugasikan reaksi carbodiimide (EDAC). Nano RIPs terkonjugasi antiEpCAM dilalukan karakterisasi berupa entrapment efficiency, ukuran partikel, potensial zeta, morfologi nanopartikel dengan Transmission Electron Microscope (TEM) serta dilakukan uji sitotoksik pada sel T47D dan sel vero. Konsentrasi optimal dari RIPs, kitosan rantai pendek dan pektin untuk memformulasikan nanopartikel RIPs kitosan-pektin adalah 0,01%; 0,01%; 1% (b/v). Nanopartikel yang dihasilkan memiliki ukuran rata-rata 376,8 ± 105,2 nm, indeks polidispersitas 0.401, potensial zeta 43,71 mV, entrapment efficiency 98,97±0,12% dan morfologi partikel yang sferis. Uji pada sel kanker menunjukkan nanopartikel terkonjugasi antiEpCAM meningkatkan efek sitotoksik RIPs pada sel yang mengekspresikan EpCAM. Dari hasil penelitian menunjukkan bahwa nanopartikel terkonjugasi antiEpCAM mampu menghantarkan RIPs secara spesifik pada sel yang tinggi ekspresi EpCAM.

Ribosome Inactivating Proteins (RIPs) are protein from plants which depurinate ribosomal RNA. RIPs isolated from Mirabilis jalapa L. leaves showed higher cytotoxic effect on malignant cells (T47D and MCF7 cell line) more than on normal mononuclear cells. Chitosan nanoparticles have frequently been used in protein delivery applications. However studies regarding protein delivery using chitosan nanoparticles modification with antibodi specifically targeting EpCAM in tumor tissues are lacking. The aims of this study were to develop RIP MJ entrapped into nanoparticles conjugated antiEpCAM, to characterized nanoparticles of RIP MJ conjugated antiEpCAM, and to study cytotoxic effect on T47D cell line. RIPs loaded chitosan nanoparticles (RIPs CS-Pec NPs) were prepared using low viscous chitosan and pectin as cross-linker with polyelectrolit complex method, then conjugated with antiEpCAM antibodi by carbodiimide reaction. AntiEpCAM conjugated RIPs CS-Pec nanoparticles was then characterized for its entrapment efficiency, particles size, zeta potential, morphology by transmission electron microscope (TEM) and cytotoxic assay on T47D and Vero cell line. The optimal concentration of RIPs; low viscous chitosan; pectin to prepare AntiEpCAM conjugated RIPs CS-Pec nanoparticles was 0.01%; 0.01%; 1% (m/v) and showed satisfactory results i.e. average particle size 376.8 ± 105.2 nm, polydispersity index 0.401, zeta potential 43.71 mV, high entrapment efficiency 98.97 ± 0.12%. Transmission electron microscope (TEM) imaging showed a spherical and homogenous structure for nanoparticles. Test on cancer cell shown that nanoparticles conjugated with antiEpCAM increase cytotoxic effect of RIPs to cell which overexpresss EpCAM. It shown that AntiEpCAM conjugated RIPs CS-Pec nanoparticles suitable to deliver protein to target cells which EpCAM overexpression .
